"If an instant ability is prevented from
activating, it can never activate, even if the prevent is later negated. If an ongoing ability is
prevented, and the prevent is later negated, the ongoing ability activates at that point." -Reg Version 5.0.0

Trembling Demon
If no good enhancements are played this battle, protect all lost souls from rescue. Opponent may discard a good enhancement from hand to negate this.

Rescue Moses, block Trembling Demon, negate Moses.. Souls are protected?

Looks like the first part of the ability (LS protection) is on-going but the discard from hand to negate it is an instant ability. So if Moses is negated I’d say that the first part still works but the second part doesn’t.

A very good question. 

This is a very unique ability, and there are very few Redemption cards like this one.  It's like TD has a built-in "Instead" ability that isn't an Instead, but acts similar to one by giving your opponent a way to Negate part of TD's ability.

And while this "Instead that is not an Instead" is an instant ability, I don't think it tries to activate unless the first ability activates, because it is tied to (and provides your opponent an "out" for) the first ability. 

So I would say that yes, they are protected, unless Moses' controller discards a GE.
