If I have four redeemed souls and rescue the lost souls card once do I win our do I have to rescue it again?

You have to rescue it again.  A half rescued 2-Liner doesn't count towards the game score.

Question: before the whole New Jerusalem ruling that made it impossible to use New Jerusalem to gain a lost soul over the max, was it possible to win T2 with 9 lost souls? (You have 6, then rescue 2-liner once, and use Son of God and New Jerusalem to rescue 2-Liner the second time and rescue another lost soul).

Yes.  You could have won with 10, if you had 6 rescued and 2 half-rescued 2/3-liners, so you could play SoG/NJ on the other two halves.
