Hello

when i discard my opponent,s "musician chamber" what happens to the cards that are in it?
dose he discard them or put the cards in his hand?

Unless otherwise specified, things that affect cards also affect the cards it holds. In this case, when Musician's Chamber is discarded, the enhancements within are discarded as well.

Yeah, things that are considered on or in something else follow that thing wherever it goes. If a character is removed from the game while holding a weapon, that weapon will also be removed from the game; if a fortress is discarded while holding enhancements and/or characters (Musicians' Chambers, Storehouse, The Darkness, etc.), all of those will be discarded also.

unless you killed it with spreading mildew which I believe says return the cards in fortress to holders hand

Spreading Mildew specifies otherwise, so it still follows the rule. It doesn't instantly discard the fortress either.
