For Isaiah angels to count toward Isaiah's identifier, do the angels need to be in play? Or can they be in set-aside? (Chamber, etc)

X = # of your Isaiah angels

As far as I know, identifiers, much like special abilities, default to in play.

However, I could also see an argument that the only restriction is "your" and cards you own and control that are in set aside are still "yours."
