I have a few lost soul questions:

Who chooses which lost soul a RA is made for when there are multiple choices, the rescuer or defender? (I understand the rules regarding access)

When a character is captured and thereby treated as a Lost Soul, if that character is rescued, does that add to the rescuer's Redeemed Soul count for the game?  And where does the character go if not, into the owner's hand or their territory?

If the defender chooses which soul the attempt is made on, and the defender has captured characters, (assuming the character rescues do NOT count toward redeemed souls total) does that become a potential deck strategy; to wit, capture characters and force the rescuer to rescue the characters to delay their rescue attempts against lost souls?

In type one person blocking.

Yes it count as a redeemed lost soul. If it isn't rescued, it just stays as a lost soul

They do count but there are strageties with capturing characters (mainly Raiders camp and demons now since demons can't be redeemed) to slow that down

So the rescued character just goes into the Land of Redemption, right?  It doesn't go back into play...

I'm assuming the benefit of capturing a character is to prevent its use if it is particularly strong, weighed against the drawback of providing a lost soul for opponent to rescue?

That's correct. There are a couple characters that are so powerful, one of the best options for dealing with them is to capture them and then rescue them with Son of God.

(That way they can't use I am Redemption, Covenant with Palestine, or any of the Shufflers to get them back)

Falling Away if possible. then other options appear- to get these "powerful" heros back. That would be a good reason to make a card that helps get them back.
