You need a gray evil character or clay hero to place the card in your territory, but do you need those brigades present in battle in order to activate its special ability?

In other words, could I use it in a battle involving a blue brigade hero?

Once you place it, it has been 'played' and therefore you do not need a brigade matching any longer. It is either a good or evil card and with a brigade based on who you played it on, but beyond that you need nothing to activate it.

Can you use it to negate thins like Dou, where you do not get special initiative?

Yes. Because it negates "last" it can negate something that has even left play (which DoU has also done) and is done anytime a manual trigger is allowed.
