Part of Nero's SA says, "Discard all Roman sites in play." Roman Prison's SA says, "If you use a Roman soldier or emperor to block a rescue attempt at this site, you may draw a card or reveal opponent's hand." If you use Nero to block a recue at Roman Prison, will you still be able to use Roman Prison's SA?

Nero's special ability completes before you trigger RP. At that time, RP is no longer in play, so I think it doesn't activate.

I think the answer to this question depends on the outcome of a discussion that was happening among the playtesters before/during Nationals. There were questions about when exactly a block is determined for the sake of resolving abilities. Some of the examples provided were:
Michael + Angel's Sword blocked by Seven Sons of Sceva who exchanges with an orange demon in the discard pile. Was Michael blocked by a human?
Priests of Christ blocked by King Zimri from hand. When Zimri's ability completes both Zimri and PoC are discarded. Does PoC still trigger the search from deck?
I think the Emperor Nero + Roman Prison situation would be resolved the same way. I don't recall that an official stance was ever posted about how to handle the above scenarios.

I think you are right.
Your second example is particularly similar, since the card with the trigger (PoC) is no longer in play, just like Roman Prison is no longer in play to conduct the check, if the check is not triggered till the abilities all complete.

The condition on Roman Prison is a static condition (it's waiting for an event to happen). The condition is satisfied (the event happens) when Nero enters battle. So the ability is carried out as soon as possible after that as long as the ability is not negated. It this case as soon as Nero's ability completes the ability of Roman Prison happens (even though at that point Roman Prison is in the discard pile).

I agree that Roman Prison and Priests of Christ should both work in their respective scenarios.

i agree also. the trigger for the ability happened, and the ability was not negated.

Yeah, because he technically was blocked by a human; that human simply exchanged with a demon because of his SA. You might be able to say that Mikey was blocked by both SSoS and the demon he exchanged with, but you still can't deny that he was blocked by a human.

Agreed. Also of significance, IMO, is that the Hero either has access or does not at the time he enters battle. So if he has access, he is making a rescue attempt "at that Site". The blocking EC may discard the Site but he does so once we've already established the condition (rescuing from the Site).

So, bottom line, is it a good idea to have both Nero and Prison in the same deck?

Depends on if you care that your Site gets killed.
