If my opponent goes out for a lost soul and I put the hard-hearted pharaoh and banned in Egyptian Warden "you May discard an evil gold enhancement from hand to capture a human hero in battle" and I discard a gold enhancement during special initiative can he negate the pharaoh who banded in Egyptian Warden to stop the capture? I thought during special initiative you were only able to interrupt and prevent what is causing the removal which would be the Egyptian worden. Right?

Correct.

Right--you could play a "negate an Evil Character card" (Samuel's Edict) to negate Warden or you could play an ItB card since that would target Warden for interrupt as the last card played (and it was played by the opponent).

Let's say Lot's Wife banded in EW which now gives SI. Wouldn't he also be able to play Samuel's Edict on Lot's Wife to cascade negate the capture?

No. If you interrupt the removal first, that would work.

Correct, you could interrupt Warden and then play something to negate Lot's Wife, which sends Warden back to territory, but you still need to interrupt Warden first.

An example would be if running Disciples. Play Reach of Desperation to interrupt Egyptian Warden, then after the draw you could play My Lord & My God to negate and discard Lot's Wife, which I believe should cascade everything else.
