What character special abilities are active outside battle? when do they activate (start working)?

for example "while this card remains in play.... blah blah blah"

Once they enter battle, the abilities activate. If the character leaves play, then the ability is no longer in effect.

Also, the ones that say "while this card remains in play" activate when they enter battle, and are ongoing for as long as that condition (being in play) is met.

If I set Esther (with activated ability) aside with Pentecost, do I have to put her into battle again after she returns to reactivate her ability?

That is correct. Setting her aside takes her out of play, so the ability resolves and is no longer active.
