Where can I find the rules for building 100+ card T1 decks?

The rules for all T1 decks are the same.

Dominants and multi-color cards are up to 1 per deck.
All other cards with SA's are 1 per 50
Cards with no SA's (except LS's and Sites) are up to 3 per deck.
LS's with no SA's are only restricted by deck size.
Sites with no SA's are only restricted by number of LS's must be greater than or equal to total number of sites.
Max deck size is 154.

I'm also looking for finer game play rules, like the rules for unique characters, etc. I've never tried to build a big deck like that, but I'd like to.

If you're wondering about how many unique characters can go in a T1 deck, the answer is 1 per deck for multi, 1 per 50 for single-brigade with SA, 3 per deck for single brigade with no SA.

If you're wondering about how many unique characters should go in a T1 deck, this thread should probably be in a deck advice forum. I would be happy to move it there for you if that's your intent.

A couple more questions that the Type 1 deck-building rules in the REG didn't cover. Can you only have one of each artifact, curse, and covenant in a deck, or is it one per 50 cards? What about Lost Souls? Sites? Fortresses?

Also, for actual game play. What are the rules on having multiples of a card in play at one time? My understanding is that you can only have one unique character in play/set-aside at a time, but as many generics as is allowed in the deck. Is that correct? What about artifacts, fortresses, sites, enhancements, etc? I looked for a little bit but couldn't find anything in the REG. Is there an article with all this information?

Sites are generic, Fortresses are Unique.  You can have multiples of Enhancements (3 Assyrian Archers each with 2k Horses?  Yes.) but I'm not sure about artifacts.

1 per 50 for each artifact/curse/cov/ls/site/fortress

I think all of this is covered in the rulebook (if not, it should be), but I'll help you out:

All cards in T1 that aren't dominants, multi-colored good or evil cards, or cards with no SA are 1 per 50. Doms and multi-colored cards (except sites) are 1 per deck. Characters and enhancements with no SA are 3 per deck. Sites with no SA are only restricted by the rule that #sites<=#LS's. Lost Souls with no SA are only restricted by deck size.

All Fortresses, Artifacts, Covenants, and Curses are Unique, so you can only have one in play or set-aside at a time. You can have as many of each generic character in play/set-aside as you have in your deck, but Unique characters are restricted to 1 at a time. Sites, LS's and enhancements are all generic.

Alright, thanks a bunch guys!

I'm a bit fuzzy though, and I may have just missed it, does that mean that I could theoretically have two Revealers and two Herod's Dungeons in a 105 deck, or not?

Two more questions, and I hope that's all.

Can I have two of one artifact in an artifact pile at a time, since that's not technically "in play"? I'm assuming not.

Also, I can have multiples of a card in my discard pile. Like, if I use Chariot of Fire, I could place two Matthews or whatever in my draw pile?

Yes to both. Uniqueness only matters in play and set-aside. Since your artifact pile (except the currently active artifact) and your discard pile are not in play, cards there do not count towards the "1 of a unique card" rule. You can use Chariot of Fire to put all copies of any specific hero back into your draw pile.
